I noticed that within 2 days after differentiation with PMA part of the macrophage-like THP1 cells underwent dedifferentiate back to the monocyte phenotype, part of them became detached and changed their morphology into rounded monocyte morphology rather than macrophage-like morphology. Since in my project I'm interested in longer term incubations, I want to characterize more accurately the time the macrophage-like THP1 stay in their macrophage-like phenotype. I want to use immuno-fluorescent stain since my cells are embedded in collagen, looking at markers used the most frequent markers are cd11b or cd36. Can cd68 be used as a differentiation marker?
