I'm facing a strange problem with my Ni-NTA pull-down.
So I transfect my HEK cells with one plasmid having a His tag (Protein tagged) and one other protein without the tag (protein B) and I'd like to determine if the two proteins interact. After transfecting my HEK cells with either the protein tagged or the protein B or both, I obtain the following results by western blot: I can observe my tagged protein when it's alone in HEK cells but no more when the tagged protein is with the protein B.
Concerning the revelation of the protein B, I observe that the staining is a little bit stronger when the two proteins are together.
So is it possible that when the two proteins interact the HIS tag is no more visible and thus the protein does not bind to the Ni-NTA beads?
