I'm trying to do an endothelial tube formation assay, but I'm having trouble with Corning's Matrigel used for this.
The CoA states that the protein concentration is 9 mg/mL.
But in the manual of Corning, it says to dilute the protein concentration to 10 mg/ml for endothelial tube formation assay... Isn't it impossible to dilute it? I looked at other reviews and used it at 1:30, 1:100, etc... I think I'm missing something, but can someone help me?

The basement membrane extract (BME) concentrations are highly variable dependent on the lot. BME does not work well for endothelial tube formation assay, if the concentration is less than 10mg/ml. Therefore you need to contact the BME manufacturer prior to purchase to ensure acquisition of an adequately concentrated lot.
The typical protein concentrations for Corning Matrigel matrix are between 8 to 11mg/mL. For endothelial tube formation assay, the Corning Matrigel matrix concentration should be at least 10mg/mL.
I suggest that you do not dilute by fold dilution, but rather dilute to a specific concentration not less than 10mg/mL. The Corning's Matrigel having protein concentration of 9mg/mL (as it is already of a lower protein concentration than required) may not be appropriate as per the manual provided by Corning for endothelial tube formation assay. So, you need to purchase Corning's Matrigel having protein concentration ≥ 10mg/mL.
