Typical function of aminoacyl tRNA synthetase (aaRS) is to attach amino acid to tRNA for following protein synthesis.
I am curious, is there a natural process (or engineered aaRS), which would attach a polypeptide to a tRNA, instead of a single amino acid?
This occurs during ribosome translation. Sometimes a peptidyl-tRNA falls off the ribosome during elongation, resulting in a free tRNA with a peptide bound to it. The enzyme peptidyl tRNA hydrolase (Pth) removes the peptide to regenerate the free tRNA.

I have never heard of wildtype or engineered aaRS that could aminoacylate peptides, but the Suga group has developed ribozymes (Flexizymes) that can attach any amino acid to any tRNA and they have shown that they can also attach peptides to tRNA using these ribozymes: Goto, Y. and Suga, H. (2009) Translation initiation with initiator tRNA charged with exotic peptides. J Am Chem Soc, 131, 5040-5041.
