i just want to know the accuracy of calcineurin antibody result and calcineurin colorimetric assay for brain tissue homogenates....and if i do western blot for calcineurin A and B subunits they both should be high? if calcineurin is activated?
Yes, if calcineurin activity is increased then definitely the increase should also be reflected in western blot. Colorimetric assays are somewhat non specific that's why fluorescence based assays are always preferred over them. Western blot is confirmatory and it will give you specific results. If everything goes fine then your colorimetric observation should reproduce into western blot results..
we have struglled with reproducability and accuracy of available assays and use both western and flourescent assays for calcineurin activity - alternative markers could be RCAN1-4 expression (downstream of calcineurin) and NFAT translocation (cytosolic or neuclear)
