I am seeing the protein expression levels of the control animal group, where I have selected 3 animals, out of which I am checking the protein expression in Muscle, Liver, heart, Adipose, Deudenum, and Pancrease, and am loading 40ug of protein on to the gel.
here my question is whether the protein expression level of Beta-actin will be the same in all the tissues....?
Beta-actin expression may vary between different tissues also between species and so on.
However, this question is not important regarding dana analysis. The most important fact in this context (utilization of beta-actn as loading control) is whether its expression is modulation by the stimulus used in your experimental setting. If there is no variation between samples (from the ame tissue) beta-actin (or any other control protein) can be used as loading control (independent from potential differences regarding its expression in different tissues or species etc.).
It is known that the basal levels of beta-actin vary with organ/tissue. Further, several stimuli, including certain cytokines and sex hormones regulate the basal levels of the beta-actin protein. Therefore, other proteins are often used to normalize the expression levels of the protein of interest.
Abdul Majid, the beta-actin expression levels should not be the same as you are looking at tissues from different organs. Like Divaker and Andreas pointed out, the experimental conditions are a huge factor and could be responsible for some variations.
However, what you can consider doing if you really want a consistent normalization for the protein samples from different tissues (regardless of the experimental conditions) is to use Total Protein Normalization (TPN) reagent. You can buy that from Bio-Rad or ThermoFischer or Azure Biosystems.
