I think there is misunderstanding, because I don't want protocol, but I've liked to know innovative information on recent progress in this concern ( regeneration from mature leaves) and available for all researchers . I need only, yes or No . Sure there are many media and I know more on plantlets regeneration from shoot tip. Anyway thanks for your advice..............................regards

My answer had been very short. And could have sounded not so nice. But it is really like that.

Normally there are no problems with getting tissue culture from a young leaf and maybe a plant still in the tube only more mature.

But to obtain a culture from a already potted plant in order to multiply it as the seeds would not be true to its genetic composition, that's something else.

So far we did not work with date palm. But just recently it succeded with a leaf from a potted plant on a medium thus far only used on young tissue.

Nevertheless you should learn all there is about the date palm. Maybe another piece of it, e.g. the stalk is more willing to colloborate. Any piece with more recent grown tissue.

My medium is MS with 1 ppm BA and 0.1 ppm IAA. Sounds so simple.

As per my recent survey for micropropagation of date palm, there is a research paper which developed a protocol from young in vitro regenerated leaf sheath part for indirect somaticembryogenesis. And that paper also says that propagation from mature leaf is quit difficult. Topic and reference of that paper is "Histological changes on regeneration in vitro culture of date palm (Phoenix dactylifera) leaf explants. Different plant species have their own preferred explants and methods for tissue culturing to achieve highe efficiency of regeneration in vitro. Other explant tissues might still can be used to induce/generate plantlets, but with low efficiency. People will tend to choose the established high-efficient method for that purpose thereafter.

Similarly in Date palm tissue culturing, I strong believe that experiments of using mature leaves for plantlets generation most likely have already been tried and compared with other explants, and come up the results that 'shoot tip' is the best one. The reason I believe so is because that among the very first things people want to know for tissue-culturing a specific species is which explants are the best for inducing plantlets.

You mentioned in your earlier post that you would like to request some new info and trend about Date palm  tissue-culturing. I attach a 2015 paper for you (see below).

There is enough literature on date pal tissue culture and you should be able to comb the literature. nmy opiion yuong leaves are totipotent and via callus one generate plants.I think this is possible.There is another route go for somatic emrygenesis .

We are  working  on tissue  culture   date  palm  since  1989 in iraq .  Unfortunately  no way   to get plantlets from  leave  or fruits.
