The most common assay among all those evaluating cytotoxicity is the MTT assay. MTT is an invitro cell-based assay which utilizes mammalian cell lines to test the cytotoxic effects of candidate drugs.

For instance, if you may want to check the anti-tumor effects of chitosan nanoparticles either Au-based or Ag-based, you could use a number of mammalian cancer cell lines as well as simultaneously you could also check the cytotoxicity of the nanoparticles in human peripheral blood mononuclear cells (PBMCs) using MTT assay.
