I have read that T-cell epitopes are all linear but have not seen any claims if found on the surface or internal part of the protein. I have been looking for references on this subject but I can still not fully answer my question. I am interested about cereal proteins. Thank you.

A follow-up question would be:
In most peptide-based gluten detection assays i.e. ELISA test kits, use of cocktail extraction releases the disulphide-bonded prolamins from glutelins in heat-processed samples, hence antibodies may bind to these native prolamins (for example, G12 mAB for 33-mer epitope). From this, can I deduce that this 33-mer or epitopes then reside in the surface of the native proteins? Thank you.
