So, there is no short answer to this as there are a plethora of agents that should work. If you are looking for a positive control, then I would recommend agents that are routinely used and published with in the area you are studying with the cell line/lines you are using. Personally, I like the vitamin A derivative, fenretinide. MJ is not necessarily correct. The differentiation-apoptosis-necrosis axis is dependent on cell line, dose/concentration, and mechanism of action of the agent.
Generally speaking biologic therapies (like Gleevec) induced apoptosis rather than necrosis as they disrupt hyperactivated pathways and restore normal cell cycle in cancer cells. Classical chemotherapeutic agents usually induced cell death through necrosis.
Regarding the intimate mechanisms and mode of action you probably never receive answer here because of space limitations.
