I have to purify protein from conditioned cell media supernatant. I wanted to test out how well I can utilise centricons in concentrating and "buffer exchanging" my protein after which I can proceed to a Ni-NTA column and then a size exclusion chromatography on an Äkta column.
I have already tested out centricons on the same protein for small volumes of supernatant (ca. 15-40ml) which worked well. However, I want to upscale now and the available method is ammonium sulphate precipitation and dialysis which takes very long, about a week.
So my question is: Does anyone know of centricons large enough, or a way to utilize available centricons on the market ( like this one: https://www.sigmaaldrich.com/DE/en/product/mm/ufc9100?context=product) to concentrate protein from a large volume of Supernatant (around 1 litre).
