I am new in doing cell death. when I treat cells with TNFa and Birinapant at different timepoint, I observed that more cell float up with longer treatment, but when I did Western Blot to detect caspase 8 and caspase 3, I observed less pro-caspase3 as well as cleaved caspase 3 with longer treatment. Will I be able to see necroptosis with TNFa and Birinapant treatment? what are exact floating cells? Are they dead cells? how can I know how many kinds of cell death after treatment? Are annexin V+/PI+ cells going through late apoptosis or necroptosis? Thank you very much in advance.
