Hi,
I recently want to do some ChIP-seq for my project with liver hepatocyte. I use the following RIPA buffer (attached picture1b) for cell lysis and sonication. And I used Covaris sonication: 16 cycles, 60sec time, Duty Factor 5.0%, Peak Power 140W, cycles per burst 200. But I didn't see DNA fragment on 1% agarose DNA gel. What do you guys suggest me to do next?

Did you decrosslinked the chromatin before the loading on the agarose gel?
Marco Vezzoli Hi Marco, thanks for your attention. Yes I did. But I used an old 1XTE to prepare elution buffer. Do you think this could be a problem?
For the elution step I use 1% SDS 0.1 M NaHCO3. Can you share your protocol?
