I have a mutant bacteria that is more tolerant to H2O2 and I would like to test if it is more persistent in the macrophage model? Will either of these cell lines phagocytose and degrade bacteria, and if yes, will it happen automatically or do they need additional factors (e.g. antibodies)?

The best would be to feed the bacteria to neutrophils and see if they can kill them.
Both macrophage cell lines should work if you stimulate them appropriately - IFNgamma together with the bacteria.
But I would otherwise use either human monocyte-differentiated macrophages or rodent primary macrophages (peritoneal or bone marrow-derived) as a cell line is always a cell line.
If I would choose between P388 and RAW I would choose P388.
I fully agree with Dr. Robert Harris's suggestions. Neutrophils are more efficient in bacterial killing becuase they can generate HOCl (bleach) from H2O2 and chloride ion using myeloperoxidase.
I fully agree with both above answers.I am also not a friend of cell lines.If possible,try to use human neutrophils or human monocytes (to get from blood banks) and gently purify them. The best way of monocyte purification has been described by us in :J.Exp.Med.184,1191-1196,1996.
I agree with Robert. I would use neutrophils - they are also much easier to work with in culture than monocytes or macrophages. We usually stimulate neutrophils with fMLP.
