Aortic ring assay is one of the mostly used method to study angiogenesis. In several publised papers, mouse aortic rings was used for this assay. While, I chose rat aortic rings from which sprouting is formed more nicely. Unfortunately, after two months' pretest, I still can not repeat the effects of a postive microRNA. Dose anybody had this experience before? If so, would you please kindly remind me of the tricky parts of the protocal. Thanks ahead!

My team also uses this assay with mouse aortae, yet I dont have experience with your specific problem. But a pretty helpful protocol has been provided by Baker et al. in Nat Protocols, see PMID 22193302. I recommend you see if that paper helps or you may directly contact that group perhaps.
