Hi all, I'm starting to use Daoy cells (medulloblastoma), and I would need to transfect them. In literature seems that there is no info on that...Has anyone tried that? thanks
Yes. I have done transfection of DAOY medulloblastoma cells several years ago using oligofectamine, but since then there have been better reagents marketed with fewer non-specific cytotoxic effects. Are you having a certain issue with transfecting you DAOY cells?
Hi Pavel, thanks..I'm just starting to do transfection in these cells, so still I have no issue..but I have either lipofectamine and CaCl2 method, and I would like to know which could work better..to save time and money...
I recently had to establish transfection of DAOY cells as well and tested CaCl2, Polyethyleneimin (PEI), Lipofectamin2000, FugeneHD, Viafect, Metafectene, Turbofect and Viromer RED.
In my hands Viavfect (1:1,5; DNA:Reagent) was the winner by far. FugeneHD and Viromer RED also worked ok, but the other reagents/methods didn't work, or were toxic for DAOY. I used 1µg Firefly/ 0,2µg Renilla Luciferase constructs per 100.000DAOY cells in 24well plate for this test and monitored luciferase activity after 42h.
thanks for sharing this experiment! I'm going to give a try to Lipofectamine (we already have it in the lab), but if it doesn't work I'll go for Fugene then...(i don't know this Viafect or Viromer RED)...thanks again for sharing!
I can also vouch for Fugene as being quite good all around even though I've never used it on DAOY cells it has worked well in various other cell lines.
