Germination seed is often a good choice for regenerating plant in vitro. But in my experiment I have failed which only ends up in some kind of colorless tissue with no any significance changes or callus, although I have added appropriate concentration callus inducing PGR.

Depends what plant you are working on may be it need low concentration of PGR.

Please check low concentration of TDZ + NAA and BA + NAA. Mostly nodal parts is giving nice results. For acclimatization use IBA. Please check these articles for help. regards and all the best,

Different plant species has different preferred tissue parts (explants) for callus induction. You need to check out some literatures to see which explants are the best for your species.
I am not able to understand your question correctly - Are you trying to grow a invitro axenic seedling, from which you propose to take explants for tissue culture? -
or you are trying to regenerate plants from part of the seed?
I think you mean the second possibility where you are trying to regenerate from a part of the seed - in that case you will not get any regeneration until you are using the embryo as other parts of seeds are storage tissue and will not respond. So try taking out the embryo and culture it.
You may try whole seed ( sterilized) germination in vitro there should not be any problem if your seeds are viable - frst germinate imbibed seeds in 1/2 strength MS medium - you will get a in vitro seedling after a few weeks and now you can use this seedling as a ex-plant source - no need to sterilized further.
it can be because of bleaching effect due to high concentration of chemicals and long time exposure your material to the sterilizing agents (streptocyclin, bavistin, HgCl2 etc.) used in pre-treatment during sterilization before inoculation.
possible approaches:
1. cut down your exposure time (more effective) i.e minimum exposure to water, before inoculation.
2. or else can try with low concentration of pre treatments.
