Many lab deprove the old nitrocellulose membrane and again incubated with new antibody. I am little confused about the new and reliable result by using the old nitrocellulose membrane .is it give the new and reliable result?
it is a common procedure to strip a membrane if sample amounts are very restricting. If you only have enough sample for one blot, reprobing is the only choice you have to analize several targets. But you have to take care of two things.1) make sure that you do not get any signal from your already analysed targetafter stripping (no antibodies left). 2) You will loose protein from your membrane during every step of your Western Blot procedure. Make sure to have a valid normalization, e.g. total protein visualization. This way you can be sure that each of your results is not influenced by protein loss.
