I am trying to perform AAV long-read sequencing using the PacBio protocol.
The protocol requires 1.5x10e12 Vg input to use 100uL and get 1ug DNA at the end of extraction, in order to get around 200ng DNA after the purification processes.
What would be the ideal extraction kit for the best yield possible?
PacBio is recommanding PureLink Viral DNA from thermofischer but I was curious to know if there were other existing kits available that could perform better?

During my PhD, I used the High Pure Viral Nucleic Acid Kit from Roche for extracting DNA from AAV vectors. Although I have never compared it with other kits, I found it to be efficient. However, it's important to note that quantification of the extracted DNA might not be straightforward. This is because the extraction kit uses carriers that will also be quantified along with the DNA. Hence, using a Nanodrop or other similar methods for quantification may not be accurate.
