I follow the below protocol to make 4OH tamoxifen in corn oil.

1) 50 mg tamoxifen + 2.5 ml of 100% ethanol (add in the same amber bottle in which Tamoxifen comes) 2) Keep the amber bottle at 37C bath and vortex occasionally until Tamoxifen is dissolved 3) Dispense 800ul of corn oil per Eppendorf (6 tubes) and then add 400ul of the ethanol-tamoxifen in each tube- 4) Vortex very well to mix and make the solution milky/cloudy 5) Open the tubes and vacuum centrifuge until ethanol evaporates (for 30 min in medium speed) Final stock concentration: 10 mg/ml

I keep these aliquotes in -20C and even after 2 days, the tamoxifen precipitates out when I thaw it. I tried heating it at 55'C for 5 mins and it does help a little but I still see particulates. I want to know if there is a better way? Also, heating 4OH tamoxifen at 55'C could cause its degradation?
