I am currently doing cell culture on Wharton -Jelly Mesenchymal Stem Cell. Is anyone know whether 10nM or 100nM of dexamethasone is the best for osteogenic differentiation.Your help is highly appreciated.
I haven't tried it with primary MSCs, but I used to use 100nM for C3H10T1/2 cells. Cells seemed to differentiate well within the first week, but when I expanded the culture period from 2-3 weeks, I observed many adipocyte-like cells, which were positive for oil red.

100nM seems to be optimal to direct MSC toward osteogenic differentiation. I was used the same concentration for hMSC towards osteogenic differentiation and quantification of Ca++ accumulation was performed by Alizarin Red staining which is very convenient in this context.
For osteogenic differentiation of MSCs, we cultured the cells in DMEM medium supplemented with 100nM of dexamethasone and we saw the evidence of osteogenic differentiation within a week.
