Currently I tried to co-culture some cancer stem cells accompanied with Peripheral blood mononuclear cells. As you know cancer stem cells` medium is serum free and we also know that PBMCs grow in FBS+ complete media. By studying I understood that there is a medium called X-vivo 15 in which we are able to culture PBMCs in the absence of FBS. I desired to know if it is possible for us to maintain PBMSc` activation in the absence of FBS or not.If yes, in this case this medium is suitable for use in here?

We have been using X-vivo for many years. it is great for routine PBMC culture without serum BUT you don't know what's in it (the recipe is proprietary). This might not matter to you, but it has become increasingly problematic for us. I have recently had it analysed by LC-MS and am rather shocked that this is not fully disclosed. I am discussing the position with Lonza right now...
