CFSE proliferation test
Dear all,
A question occured to me that if CFSE application is suitable for testing proliferation of T cells exposing to IL-2 or PHA for 14 days?
For flowcytometric anlysis on day of 14, are the untreated and CFSE dyed T lymphocytes viable as a control?
...it's probably unlikely that you will have a decent fluorescent signal remaining after 14 days, if at all.

yes its possible to expose you cell to IL-12 ,IL-2 or...any stimulants for up to 7 day in the peresent of CFSE dye.

You can do it by 5 days treatment too. So many articles are available in PubMed.

Generally, 14 days is quite long to retain the CFSE signals, however it also depends on how fast your cells are proliferating. I would give it a shot by loading CFSE at higher concentration (10 uM) to maximize the range of CFSE dilutions as much as you can.
