Is it possible to performed X-ray diffraction (XRD) analysis of protein powder without preparing crystal of protein. Is the diffraction data for protein can be use for secondary structure analysis. Is it possible to analyse protein interaction with surface metal and can say something about the interface or lattice point of a metal crystal that interacting with the protein.

yes, it is possible to perform x-ray diffraction analysis of protein is in powder form by using high quality powder X-ray diffraction techniques. but it needs a high intensity 2D detectors.the protein powder diffraction data can't be useful in secondary structure analysis. u need circular dichroism spectra for secondary structure analysis.
plz u can search literature regarding to protein interaction and lattice model.
Small angle x-ray diffraction (SAX) of the protein in solution may help characterize the protein in the absence of crystals. This method can give a sense of shape and oligomerization state of the protein.
