I am looking for a way to quantify my exosomes after I have isolated them. I do not have access to a Nanosight (NTA) and although I can do EM this wont let me to quantify them.
Would you recommend or do you have any experience with EXOCET assay which rely on the Acetyl-CoA Acetylcholinesterase? They said this enzyme is enriched on exosomes however they just give two papers backing it up

I recommend for you ExoELISA (System Biosciences) because NTA and electron microscopy have not affordable.

I sugget to use ExoTEST from HansaBioMed. It's a double sandwich ELISA which includes exosome standards for calibration curve designing. Compared to other quantification kit it is the only which allows a specific capture on ELISA plate based on immunoaffinity.

In my opinion the only way to quantify exosome Particules consists in using nanosight but when it's not possible (like in my lab) you can use ExoTEST or ExoELISA or westernblot quantification of Flotilin, CD63, CD9, CD81 etc....but be careful because markers depend of the cell origin of exosomes. and if you want to treat cells why exosomes you have to take more care about the method to obtain your exosomes (number of cell seeded, cultures times, volume of culture supernatant etc..)
