I cultured sheep ovary fragments for 7 days and after that they were fixated in Carnoy´s solution(ethanol 100%, chloroform and pure acetic acid 6:3:1) for 4 hours and conserved in ethanol 70% until paraffin embedded. Some histological sections show signs of autolysis. I intend to do immunofluorescent staining for proteins that are in the cell nucleus. Will the tissue autolysis interfere (in a negative way) with the immunofluorescent staining by altering the results or not detecting the proteins properly?
Depends on the degree of "autolysis" (I am not sure how you judged) but yes, generally if the tissue starts to decompose, any staining results of that tissue should be interpreted with caution. You can still proceed with the experiment and see if it works - it might.
