I cultured sheep ovary fragments for 7 days and after that they were fixated in Carnoy´s solution(ethanol 100%, chloroform and pure acetic acid 6:3:1) for 4 hours and conserved in ethanol 70% until paraffin embedded. Some histological sections show signs of autolysis. I intend to do immunofluorescent staining for proteins that are in the cell nucleus. Will the tissue autolysis interfere (in a negative way) with the immunofluorescent staining by altering the results or not detecting the proteins properly?
