I recently conducted a WST-1 assay on iPSC-derived cardiomyocytes and encountered unexpected results. I seeded cells into a 96-well plate at three different densities (Each sample was twice the density of the previous one). Using 10ul WST-1 reagent in 100ul medium, I expected absorbance values to change with cell density. However, despite the substantial variation in cell densities, the absorbance values at 450nm were the same. I thought maybe it was because of the incubation time but when I tried to incubate them for 6.5 hours, the absorbance values were increased but, again, they were all the same for different cell densities.
I’m seeking insights into potential explanations for this outcome. Any suggestions you can share would be greatly appreciated.
