Hello everyone, I'm trying to extract proteins from J774A1 cells after treating them with 200ng/mL LPS for 4 hours followed by 5mM ATP treatment for another 4 hours to study inflammasome activation and pyroptosis. After ATP treatment, most of the cells are detached from the cell culture plate. I have tried increasing the amount of cells seeded in the culture plate but it doesn't yield much proteins after extraction. What are the other possible ways to increase protein quantity for western blots?
