I am currently working with a specific type of cells called mouse alveolar macrophages (MH-S) in my experiments. During cytometry analysis, I consistently observe two distinct populations when I separate the single cells, which is also reflected in the sharp intensity of these two populations. Despite my efforts to investigate the cause, I consistently obtain the same result. Since these cells grow in both suspension and adherent forms, I attempted to separate the populations and determine if the two populations are due to different cell types (suspension and adherent). I reduced the cell density and tested different conditions, but I am unable to comprehend the nature of the second population.
