Hi, I have been having this issue for a while now.
When I use the standard miltenyi CD8 microbeads for MACS separation, I always get a 'tail' that no matter what I do, I cannot get rid of them...what are these CD8dim cells?
things that I have tried
1) incubate beads on ice for 15 min to reduce non-specific binding
2) before isolating CD8, I depleted NK cells using CD56 beads
but those CD8dim cells are still there! What are they??

I guess you are isolating CD8 T cells from Peripheral blood lymphocytes. This type of dim marker expressing cells in the purified fraction are also observed in other isolation such as CD4 and Treg cells. The incubation at RT and reducing the time may be for 5 to 10 min may improve the results

I missed the beginning of the discussion but if you speak of CD8 dim T cells, you have first gamma delta T cells, on part of which can express CD8 aplha alpha. A minority of mucosal TCRalpha beta T cell can also express low levels of CD8 (alpha alpha homodimer and are detectable in peripheral blood. ANd of course you have some CD4+ T cells that can express Dim level of CD8.
