I am digesting a concentrated body fluid sample with proteinase K, I was wondering if anyone know how to determine if the digestion is complete?
I only know by experience that usually after digestion the solution will turn clear. Is there any other more 'scientific' way to determine it?

The inclusion of a fluorogenic protease substrate in a mock reaction is often a useful method to gauge proteolysis in complex mixtures. Thermo has several useful substrates:

When the fluorescent signal asymptotes out you can infer that the original sample is digested. An important control is of course the fluorogenic protease substrate minus the complex mixture.
