There are other posts addressing this. However, the use of STZ is tricky. Fasting or not makes a big difference so an overnight fast before STZ injection improves reproducibility significantly. Also STZ is very unstable so it should be made up fresh immediately before injection. If a significant number of animals are to be treated it may even be necessary to make a fresh batch up half way. Reason I'm asking this is I've noticed tremendous difference in rats' FBG when STZ is injected after 6h fasting and STZ injected 24h fasting. why do you think is so?
A 6h fast is not sufficient to ensure that starving has been effective. I think this most likely explains the difference. We always used a 16h fast (overnight) and were able to get consistent results.
