We have a problem with MIN6. I don't know certainly the number of passage that they are. The first passage that we have has an appropriate reaction on adding glucose, but the fourth does not answer on glucose addition. I mean, intracellular calcium level does not increase after glucose adding.
MIN6 are cultured with high glucose DMEM supplemented with 10% FBS, 50 microM 2-mercaptoethanol and antibiotics. Medium is changed every 2-3 days.
