In our investigation we faced with problem of very low concentration of nitrates in forest soils and want to find adequate method for analysis big number of samples. We are considering to use microplate reader but we don't know adequate methods for this measurements. I really appreciate if someone suggest me the procedure.
If the concentrations are low you may struggle for sensitivity with the short light path of microplates. So before you get too excited try measuring the absorbance for a known path length. The rest is calculation.
What is the concentration of ammonium and nitrate in the soil? I can suggest some things but need to know how low is it... Have you considered using FTIR-ATR? you can detect both ammonium an nitrate in concentrations as low as a few mg/L of solution with a standard FTIR. The new ones can maybe go lower than that. You can also use autoanalyzer for that...
