I'm interested in the relationship between HDAC (histone deacetylase) inhibitor (HDACi) and sepsis.
With my recent experiment, I got different patterns between mRNA and ELISA as a result of some cytokine.
My results:
1. mRNA expression
sham : HDACi : sepsis : sepsis+HDACi = 1 : 4 : 2 : 1
2. ELISA expression
sham : HDACi : sepsis : sepsis+HDACi = 1 : 1 : 2 : 1.5
How do I make a discussion with these results?
First, I think there is no problem with my supposition that HDACi has anti-inflammatory properties via suppression of cytokine protein expression because it is an active form of cytokines that was detected ELISA.
But what happened to the inceased mRNA expression in HDACi group?
Any suggestions?
Ok,this is possible there may b the role of transcriptional factors involved in signalling cascade in supression of the protein expression or alternative splicing thus mature protien as per your targetted anti-body against antigen may b low and some other isoform may b there or some cross contamination during elisa binding may b in existence.
