To create a cell line overexpressing a specific microRNA, I'm planning to synthesize and clone the shRNA sequence into a vector. Would it be sufficient to add only the termination sequence (TTTTT) to the end of the pre-miRNA sequence? Or are there any additional sequences that need to be added separately?

For example, when cloning the miRNA (pre-miRNA) DNA sequence, such as ATGACTGATTTCTTTTGGTGTTCAGAGTCAATATAATTTTCTAGCACCATCTGAAATCGGTTAT, into a vector:

Restriction Enzyme Site - ATGACTGATTTCTTTTGGTGTTCAGAGTCAATATAATTTTCTAGCACCATCTGAAATCGGTTAT TTTTT (termination sequence) - Restriction Enzyme Site

Would cloning it like this be sufficient to induce overexpression of the miRNA upon transduction?
