Hello. I've been doing qPCR of exosomal miRNA from serum and plasma. cycle threshold of sample from plasma always shows lower value than serum sample, so i can say that the level of exosomal miRNA from plasma is higher than exosomal miRNA from serum sample. I searched several journal for what can cause this, but mostly explained the higher level of miRNA in cell-free plasma sample compared to cell-free serum sample, not higher level of exosomal miRNA plasma sample compared to exosomal miRNA serum sample. Can anybody give link to journal or explanation as to what may cause this difference? Thank you very much

I am not aware as to any literature on this, but I have a guess that it might have to do with losing exosomes (and therefore exosomal miRNA) in the process of removing the clot from whole blood that may be trapping vesicles and leading to loss of sample. Whereas in the plasma there is less chance of losing vesicles and would result in higher yield of exosomal miRNA. I have only done miRNA work with cell culture supernatant derived exosomes so this is only my best assumption.
I agree with Phillip, if you try to isolate exosomes from the same volume of plasma and serum (the same donor), you will get about a half less amount from serum. Last week my experiment with plasma clotting and consequent exosomes purification has shown the same result.
I agree with Phillip about losing exosome upon clot formation. However, you have to consider that during the process of clot formation, platelets get activated and secrete exosomes that are not naturally in the blood stream. Therefore, any higher/lower miRNA expression might be the result of the process.
