Why do I get a large number of proteins post PI-PLC treatment of sperm?

I have treated 100 million spermatozoa with PI-PLC (2U/ml) in sp-TALP medium.The enzyme has specificity for GPI anchor. However, post MS analysis by TPP, approx 300 proteins are identified many of which are not even GPI linked, rather are intracellular. Why this anomaly?

Hi Vipul, all cells shed membranes (and proteins) during any type of handling. It is impossible to avoid.

I think the beast you can do is to have a control of mock PI-PLC treated cells and compare and contrast them to real PI-PLC treated cells.
