Hi, I am culturing B16-F10 melanoma cells. Usually we passed cells in every 4/5 days. But using same lot (newly thawed), cells are growing very fast and we need to passage every 2 days. Looking for your suggestions if someone face the same problem. Our target is to keep 60-80% confluency during cells preparation for injection (in mice). Thanks in advance!

B16-F10 is the most proliferative cell line among the B16 cell sub-lines. Studies have shown variable doubling time for B16-F10. One study has found the doubling for B16-F10 to be 20.1 hours, while another study has found it to be 14.2 hours.
These variable values found by different research groups is because of different chosen compositions of the growth media. Serum is the most widely used growth supplement for cell culture media. Changing the optimal concentration of serum provided in the protocol would be directly proportional with changes in cell proliferation.

you need to check the concentration of your culture medium. The more concentration % the fast will be the growing charges. Normally I used 10% FBS with normal growth to achieve 80% confluency with in 4 days.
