I'm going to create contig from meleafris EST datast, searching in est file shows that both there are both poly (A) and poly (T) sequences in EST file. Is it related to the methods used by scientists?
It is either an RNA or cDNA representation: PolyA at the tail 3' end indicates a eukaryotic mRNA sequence (except histones,18S RNA, and other nuclear RNAs), while a PolyT at the beginning (5' end) merely represents the cDNA (complementary sequence thereof). Convention has it that Uracils (U's) are typed in as Thymidines (T's) for both nucleic acid types/representations in order to simplify things... although it is understood that RNA has "U's instead of T's from Biochem ...
