I prepared the plant tissue (small cotyledones of A.thaliana or also marine microalgae Pterosperma cells) for TEM using standard chemical fixation. After osmication (3h for plants, 1h for microaglae, both at +4C), the samples were slightly dark with visible green colour. The TEM micrographs of plants were good, but the microalgae were very bright even though the ultrastructure was fine.

Is it connected to short primary fix or insufficient washing after aldehydes, or stainig with osmium on ice?
