In the biological samples (suspensions or plant tissues) processed by PLT method I experienced creation of gel-like solution close to the sample when I was infiltrating the samples with mixture of ethanol-lowicryl (ratio 1:2) at -35C.
The gel-like solution was hard to mix and dissolve in ethanol-lowicryl solution and also the solution changed from clear to mild-milky one.
Is it possible that proteins were extracted during the infiltration with lowicryl and released to solution? Or what else could be the problem and how to overcome it?

You can control the structuring of the mixture of ethanol-lowicryl. Proteins are not extracted by lowicryl.

Thank you for your respons, it helpt. I added one extra dilution step between full EtOH and EtOH: Lowicryl 1:1 ratio, and it worked out very nicelly!
