The study I am working on requires isolation of lymphocytes and neutrophils from human blood. For the separation, I am using Polymorphprep. When I stain the neutrophil-rich PMN fraction, I do not see any CD16 +ve cells, this is strange as neutrophils should stain for it. I know that these cells are neutrophils as I also perform DAPI staining and am able to see the multiple nuclei, characteristic of neutrophils. Additionally, the neutrophils are also staining for CD3/CD19 which are T and B cell markers respectively. Has anyone encountered this situation ? I would be really grateful if you could share your experience.
