I am doing western blots to detect a particular protein in crude worm extracts of a tropical nematode using antibodies raised in rabbits against a peptide from this protein. Unfortunately, the pre-immune serum seems to be giving very strong signals (unspecific) whereas the antibodies are giving a rather faint signal.
I used the primary antibodies at 1:2000 dilution and secondary antibodies at 1:5000 and each of the incubations was for 2 hours at room temperature. Blocking was with 5% milk over night at 4°C and revelation was done using DAB in 0.1M imidazole, pH7.0. The molecular weight expected is about 58.8 kDa.
