Why is neccesary or so important to have a virus titre of 100 TCID50/well when you want to perform a virus neutralization test?
I have been trying to found an explanation in the literature but I found nothing. Everybody just say: "the quantity of virus is 100 TCID50/well, no more explanation no references...
I would appreciate any help.

100 TCID50 / wel; because it is a convention (the development of the technique was like that) it is like the case of the 4 hemagglutinating units in the technique of the inhibition of hemmaglutination( HI test) for the hemagglutinating viruses as Avian influenza.
Probably because WHO recommended it decades ago for polio NT (limits: 50-200). In fact, the most sensitive assay is accomplished with the lowest possible TCID50. so, to also keep WHO happy go with 50-60.
