I was culturing the pre adipocyte -3T3-L1 two weeks ago and I found that the status of my cells died so much. When the cells disappeared there was a large amount of black dots in the culture medium which take the brownian movement. I have tried to use the antibiotic, anyway, it wasn't useful. Who knows what they are? How do I get rid of them?
Sounds like microbial contamination. Clean your tissue culture incubator (wash and autoclave the shelves), and throw out your culture medium. Make new medium and place in a dish without cells in your incubator. If the medium stays clear for two days, then you can be sure you have a sterile environment. If the contamination occurs again, then you might need to obtain your 3T3-L1s from another source, since the parental culture/stock could have been contaminated.
Are you using Penn/strep in your standard culture medium?
I agree with everything Daniel Cohen said. i would like to add that you should make sure to remove and clean the fan in the incubator. Many folks will remove all of the shelving for autoclaving and disinfect the inside of the incubator but often forget about cleaning and disinfecting the fan. If not cleaned it will re-seed the incubator with bugs resulting in contamination of your cultures.
