i suggest you should first look for TLR4 surface expression on your cells before you start your LPS-experiments. In our lab we use LPS from Invivogen which is purified from Porphyromonas gingivalis (LPS-PG ultrapure, Invivogen, Cat.: ttrl-ppglps). It is described by the authors to be predominantly binding to TLR4. We also used more unselective LPS from Sigma, but the results are not that good. For the ultrapure LPS from Invivogen we get much clearer results reflecting TLR4 signalling.

I agree with the comments above. In our hands, LPS from different sources works differently in THP-1 cells. Therefore, it is important to first determine whether cells do or do not express detectable levels of TLR4 before trying LPS from different sources.

I think you should try different types of LPS, because LPS strains work differently in human vs mouse. I am not sure in liver cells, but you might see the same.

Thank you all for the kind responses. I agree that it would be good to start with testing an LPS whose effects are transmitted through TLR4, and then work on some other LPSs when appropriate. We have our prelim results coming out, and they look fine.

Thank you Dr. Christian Müller. Our lab happens to work on Pg LPS too, through collaboration with a local dental professor. It's an interesting model.

In my experience, we use in our lab, LPS from E. Coli obtained by ethanolic fraction (Sigma. For us, it is which causes a stimulation of 9 - 11 fold vs non-stimulated THP-1 macrophages cells. Even in THP-1 monocytes is effective the stimulation. I hope that help you.
