While some experts have doubted hydroxyl radical even exists in cells, some say it does but at nearly immeasurably low amounts in extremely short time.
In your opinion, what have been the most "acceptable" or plausible cell models that demonstrate hydroxyl formation? (any classical or recent good ref?)
What cell types, what inducers, what scavengers, etc. would be useful for studying this radical in cell models?

Anything that compromises the mitochondrial permeability transition pore (MPTP) will induce the formation of hydroxyl radical - an excess of H2O2 will be the precursor to hydroxyl formation.
The hydroxyl radical react with almost all organic molecules with a diffusion rate constant. This means that it reacts with whatever molecule it collides first and its life time is extremely low. Since one radical begets another radical, the hydroxyl radical generates a cascade of radical reactions. Therefore, you can't design any plausible cell models that proves the hydroxyl radical formation. For the same reason the scavenger of HO(.) does not exist in vivo or in the model cell

The hydroxyl radical is formed by Haber-Weiss and/or Fenton reaction.
Fenton reaction: iron (II) is oxidized by hydrogen peroxide to iron(III), forming hydroxide anion and hydroxyl radical.
Fe2+ + H2O2 → Fe3+ + OH− + •OH
The Haber–Weiss reaction generates hydroxyl radicals from superoxide anion and hydrogen peroxide.
•O2− + H2O2 → •OH + OH− + O2
The Haber-Weiss reaction is very slow, but it is catalyzed by iron. This process is a cycle that starts when the ferric cation is reduced to iron (II) by superoxide anion.
