I am planning to extract cell secreted vesicles from different type of cell cultures. There are some kit based methods available. However, I am wondering if the classical differential centrifugation methodology is still the best to use.

Total exosome isolation reagent works great for cell culture. Ultracentrifugation protocols work well too, especially with sucrose gradient; they are more time consuming and tricky though, and one can not process more than 6 samples at a time, due to rotor limitation
Dear, it might not be possible to develop a universal method for EV isolation but the available standard methods applicable towards solving particular types of problems should be developed and approved. Development of such methods for EV isolation aimed at applying them in diverse scientific and clinical studies is currently a high priority task.
