As there are many hundreds of different saponin structures in plants, would you consider a colorimetric reaction and running on spectrophotometer, as you stated you would like an estimate?
i have 60 Aloe vera genotypes and want to estimate the total saponin content in each genotypes by using HPLC. which saponin standard to be used for total saponin estimation??
OK, I searched Medline for aloe saponin and got about 13 hits. Some of them are free full text papers. I did not read them.I do not know what the major saponin is in aloe. If you want to move the project along that is an important bit to find out. Whatever standard you choose, you would then express your estimation as "amount saponin as standard X". This is also why I advised that you may want to put the hplc aside and try a colorimetric approach, and look at specific reagents like Liebermann-Burchard that are known to specifically react with saponins, b/c if you do not know what saponins are in there, how do you know which saponin standard to obtain?
I am working on saponines analysis and according my experience the HILIC mode of UPLC is a convenient tool to assess the triterpenoid saponines content in extracts. The detection technique is also problem on account of no UV activity of these compounds - ELSD and especially MS in negative mpde are decisions.
